The nitrate reducing capacity of pure cultures of Cenococcum geophilum (Sow.) Ferd. & Winge, Paxillus involutes (Batsch: Fr.) Fr. (strains 1 and 2), Piloderma croceum Erikss. & Hjortst., Suillus variegatus (Fr.) O. Kuntze (strains 1 and 2) and an ectendomycorrhizal (E-strain) fungus was measured using an in vivo nitrate reductase (EC 1.6.6.3) assay. Differences between species and strains were established. The nitrate concentration of the culture medium influenced the nitrate reductase activities of the E-strain fungus and one strain of S. variegatus. The nitrate reductase activity of certain species and strains was a function of nitrate concentration. Addition of ammonium to the growth medium did not have any significant effect on the in vivo or in vitro nitrate reductase activity. The in vivo nitrate reductase activity in the mycelia of C. geophilum and the E-strain fungus decreased during 28 day growth in modified Melin-Norkrans medium. For mycelia of Paxillus involutus, Piloderma croceum and S. variegatus grown on agar the in vitro assays showed higher nitrate reductase activity than the in vivo assays.  相似文献

Body water content was determined on groups of Cyprinodon variegatus after sequential acclimation to ambient salinities in the range from fresh water to 100%. A multiple linear regression model applied to the data included body mass ( M ) of individuals and acclimation salinity ( S ) as independent variables, and body water content ( W ) as the dependent variable. The model that described the relationship was W (g) = 0·059 + 0·723 M (g)–0001 S (%). Converting values of body water content to percent of wet body mass produced a range of values from 74·5% in fresh water to 72·0% at 100%, only a 2·5% difference in body water content over this wide range of ambient salinities.  相似文献

Host plants exhibiting insect resistance traits have long been known to influence within‐plant distributions of pests and their natural enemies. Sites and timing of egg deposition are particularly important for synchrony of predators and their prey in the field. Temporal and spatial distribution of eggs of the cotton bollworms [Heliothis virescens (F.) and Helicoverpa zea (Boddie) (Lepidoptera: Noctuidae)] and that of the predators Geocoris punctipes (Say) (Heteroptera: Geocoridae), Chrysoperla rufilabris (Burmeister) (Neuroptera: Chrysopidae), and Micromus spec. (Neuroptera: Hemerobiidae) were determined during three cotton seasons, from 2002 to 2004, by collecting and examining plants throughout each season. Comparisons also were made between Bt and non‐Bt cotton to investigate possible changes in oviposition behavior on Bt cotton. The study was conducted in commercial fields with insecticide use to manage pests when economic thresholds were exceeded in both cotton types. Egg densities for predators and bollworms varied among years, but were similar on Bt and non‐Bt cottons. Oviposition of bollworms and G. punctipes correlated spatially within plants, with most eggs laid on structures in the top five nodes of cotton plants and on the three outermost leaves on lateral branches regardless of cotton type. Bollworm oviposition dynamics exhibited two peaks within the season (early July and early August). Eggs of all predators and bollworms collected from the field and incubated in the laboratory had high hatching rates throughout each season (74–100%). Temporal association of predator with bollworm oviposition showed a significant correlation with green lacewings, a delay of 10 days for big‐eyed bugs, and no correlation with brown lacewings. Furthermore, Bt cotton plants exerted no significant effect on temporal or spatial patterns of oviposition of bollworms or the predators, indicating no change in oviposition behavior of bollworm females within plant structures after almost one decade of widespread planting of Bt cotton.  相似文献

A field trial was conducted using an oil formulation of the entomopathogenic fungus, Metarhizium flavoviride Gams & Rozyspal (Deuteromycotina: Hyphomycetes), to control the variegated grasshopper, Zonocerus variegatus L. (Orthoptera: Pyrgomorphidae). The influence of dispersal and secondary pick-up from the spray residue on mortality was assessed by sampling insects from the treated plots at intervals and monitoring disease levels during subsequent incubation in the laboratory. The cumulative mortality curves showed the pathogen to be highly effective, even in samples taken 8 days after spraying. A model was developed to link the influence of mortality, incubation, secondary pick-up of spores and insect dispersal, on the shape of these cumulative mortality curves. The model proved useful for data received from an experiment using small plot sizes, where insect dispersal is a big problem. The spray residue had an important influence on overall insect mortality. The model also indicated a difference in speed of kill between field and laboratory samples, suggesting an incubation delay in the field.  相似文献

Phytoplasmas belonging to the 16S rDNA subgroups IB and IC were found in five cyclamen (Cyclamen persicum L.) plants showing virescence and yellow stunted leaves and one plant showing phyllody, rolled and thickened leaves, respectively. Two cyclamens, representing the two syndromes, were chosen as source plants for transmission trials in which three leafhopper species, known as vectors of IB and IC subgroup phytoplasmas, were used to inoculate cyclamen and periwinkle [Catharanthus roseus (L.) G. Don] test plants. Out of 366 tested plants only one periwinkle exposed to Euscelis incisus was found harbouring a 16Sr‐IB phytoplasma. Out of 60 tested vector insects, only one adult of Macrosteles quadripunctulatus and two of E. incisus fed on 16Sr‐IB source cyclamen gave a positive amplification signal in nested PCR. This extremely low level of transmission to both cyclamen and the very susceptible periwinkle strongly suggests that cyclamen, commonly found infected in crops, is an unsuitable species for phytoplasma acquisition and can be regarded as a dead‐end host plant for phytoplasmas belonging to both IB and IC subgroups. Indications for glasshouse management are drawn from these findings. Among the leafhoppers investigated E. incisus falls most under suspicion since it feeds better than the others on cyclamen, was able to transmit the disease to one periwinkle plant, and IB phytoplasmas were detected in two individuals.  相似文献

The role of natural enemy diversity in biological pest control has been debated in many studies, and understanding how interactions amongst predators and parasitoids affect herbivore populations is crucial for pest management. In this study, we assessed the individual and combined use of two species of natural enemies, the parasitoid Aphidius ervi Haliday, and the predatory brown lacewing Micromus variegatus (Fabricius), on their shared prey, the foxglove aphid, Aulacorthum solani (Kaltenbach), on sweet pepper. We hypothesized that the presence of intraguild predation (IGP) and predator facilitation (through induced aphid dropping behaviour) might have both negative and positive effects on aphid control, respectively. Our greenhouse trial showed that overall, the greatest suppression of aphids occurred in the treatment with both the parasitoid and the lacewing. While the combination of lacewings and parasitoids significantly increased aphid control compared to the use of parasitoids alone, the effect was not significantly different to the treatment with only predators, although there was a clear trend of enhanced suppression. Thus, the combined effects of both species of natural enemies were between additive and non‐additive, suggesting that the combination is neither positive nor negative for aphid control. High levels of IGP, as proven in the laboratory, were probably compensated for by the strong aphid suppression provided by the lacewings, whether or not supplemented with some level of predator facilitation. For aphid management over a longer time scale, it might still be useful to combine lacewings and parasitoids to ensure stable and resilient aphid control.  相似文献

Interactions between saprotrophic and ectomycorrhizal fungi have been largely ignored, although their mycelia often share the same microsites. The mycelial systems show general similarity to each other and, although the enzymatic potential of the saprotrophic fungi is generally considered to be higher, the importance of organic nutrient sources to ectomycorrhizal fungi is now widely accepted. In the experiments described here, nutritional interactions involving transfer of elements from one mycelium to the other have been monitored dynamically using radioactive tracers and a non-destructive electronic autoradiography system. Microcosms were used in which mycelial systems of the ectomycorrhizal fungi Suillus variegatus and Paxillus involutus , extending from Pinus sylvestris host plants, were confronted with mycelia of the saprotroph Hypholoma fasciculare extending from wood blocks. The fungi showed a clear morphological confrontation response. The mycorrhizal mycelium often formed dense patches over the Hypholoma mycelia. Up to 25% of the 32P present in the Hypholoma mycelium was captured by the mycorrhizal fungi and translocated to the plant host within 30 d. The transfer of 32P to the saprotroph from labelled mycorrhizal mycelium was one to two orders of magnitude lower. The significance of this transfer as a 'short cut' in nutrient cycling is discussed.  相似文献

Conidia of Metarhizium flavoviride grown in diphasic culture with sporulation on rice, were extracted in kerosene and formulated with peanut oil. The formulation led to high mortality when applied to field populations of Zonocerus variegatus; between 70 and 95% of insects in samples collected from the field 0, 3 or 7 days after spraying died. Sporulation of M. flavoviride was observed in 70–80% of the dead‐treated insects, which compares favourably with results obtained in laboratory bioassays. Many dead Z. variegatus, some with internal sporulation, were found in shaded niches and oviposition sites in the treated plots. No sporulation was observed in samples collected prior to application, and both sporulation and mortality differed significantly between control and treated plots. Some infections were observed in insects collected 16 and 23 days after treatment; these late infections may be a consequence of contact with spores surviving in the environment.  相似文献
